1154 Sonophoresis enhances the skin penetration of 5- aminolevulinic acid: A promising pretreatment for photodynamic therapy

Background: 5-aminolevulinic acid (5-ALA) has a poor penetrance of the skin with topical application, which reduces efficacy Photodynamic therapy (PDT). Sonophoresis involves use sound waves or ultrasonic energy to enhance transdermal delivery drugs. The purpose this study was investigate effects sonophoresis on penetration 5-ALA into skin. Methods: We calculatedin vitrotransdermal accumulation ALA, and fluorescence images were collected for analysis. Results: cumulative amount that penetrated increased over time significantly larger than without sonophoresis(P<0.01). With 5% sonophoresis, distinct localization 5-ALA-PpIX in sebaceous glands started appear 30 min after is much earlier only. For all incubation times, intensities distributing higher treated non-sonophoresis (P < 0.05). Conclusions: could be technique choice enhancing production 5-ALA-induced PpIX improving 5-ALA-based PDT, may decrease treatment time, lower cost clinical improvement, allowing many more patients treated.
